What is FOXO4-DRI??
FOXO4-DRI (also called FOXO4-D-Retro-Inverso or FOXO4 p53-Interaction Domain Peptide) is a D-retro-inverso stapled peptide designed to antagonise the interaction between FOXO4 and p53 in senescent cells. In non-senescent cells, p53 localises to mitochondria to trigger apoptosis when activated. In senescent cells, FOXO4 sequesters p53 in the nucleus, suppressing this apoptotic programme and enabling senescent cell survival (SASP). FOXO4-DRI disrupts this FOXO4–p53 interaction, restoring nuclear p53 export → mitochondrial translocation → apoptosis selectively in senescent cells — without affecting normal proliferating cells.
